Figure S2. Cellular immune response in the local draining LN after induction of the DTH response. Draining LNs (popliteal and inguinal) from three mice were pooled, counted, and analyzed by flow cytometry or used for in vitro restimulation. (A) DT-treated DEREG mice have an elevated number of total cells, including CD4+ and CD8+ T cells, compared with DT-treated WT mice. If at all, only minor differences were observed between CD25+Foxp3- cells and Foxp3+ cells. (B) No apparent differences were observed in the frequency of CD4+, CD8+, and CD25+Foxp3- cells when DT-treated DEREG mice were compared with DT-treated WT mice. However, a drastic reduction in the frequency of Foxp3+ cells was observed in DT-treated DEREG mice. (C and D) LN cells from DT-treated DEREG mice display increased IFN-γ production (C) and proliferation (D) upon restimulation with NP-O-Su-modified DCs compared with DT-treated WT mice. Cytokine production and proliferation was assessed after 4 d in culture. Data show the mean of four samples.
