Figure S1. Monocytes accumulate in the myocardium after MI. (A) Cell suspensions from C57BL/6 mouse peripheral blood, spleen, healthy heart (Pre), or infarcted heart 1 d after MI were stained with anti-CD11b, -CD90, -B220, -CD49b, -NK1.1, -Ly-6G, -Ly-6C, -F4/80, -I-Ab, and -CD11c mAbs. Representative dot plots depict monocytes/macrophages/dendritic cells identified as CD11bhi (CD90/B220/CD49b/NK1.1/Ly-6G)lo. These cells were further divided into Ly-6Chi monocytes (Ly-6Chi [F4/80/I-Ab/CD11c]lo), Ly-6Clo monocytes (Ly-6Clo [F4/80/I-Ab/CD11c]lo), and macrophages/dendritic cells (Ly-6Clo [F4/80/I-Ab/CD11c]hi). (B) Representative cytospin preparations depicting Ly-6Chi monocytes, Ly-6Clo monocytes, and macrophages/dendritic cells after cell sorting of splenic Ly-6Chi (F4/80/I-Ab/CD11c)lo, Ly-6Clo (F4/80/I-Ab/CD11c)lo, and Ly-6Clo (F4/80/I-Ab/CD11c)hi cells, respectively. Bars, 10 μm. (C) Cell suspensions from C57BL/6 mouse healthy heart (Pre) or infarcted heart 1 or 4 d after MI. Representative dot plots depict CD11bhi (CD90/B220/CD49b/NK1.1/Ly-6G)lo and show F4/80, I-Ab, or CD11c expression separately for Ly-6Chi and -6Clo cells.
