Figure S1. mAb characterization. (a) M, molecular weight standards; lane 1, empty αDEC (αCD205) mAb; lane 2, αDEC-LACK mAb; lane 3, 33D1-LACK mAb; lane 4, Ig-LACK. Antibodies were analyzed by SDS-PAGE under reducing conditions and revealed by Western blot with horseradish peroxidase-conjugated antibody to mouse IgG1. (b) M, molecular weight standards; lane 1, soluble LACK protein. The molecular weight of LACK protein was determined by SDS-PAGE and staining with Coomassie blue. (c) αDEC-LACK binds to stable DEC-transfected CHO-S cells. 10⁵ CHO-S cells were incubated for 20 min on ice with graded doses of αDEC-LACK. Binding was determined with anti-mouse IgG1 mAb and is similar to what is observed with αDEC without a fusion protein. (d) αDEC-LACK did not bind to parental CHO-S cells, and (e) Ig-LACK did not bind to DEC-transfected CHO-S cells.
