Figure S3. SDF-1α is targeted to α-granules of the megakaryocytic lineage. The megakaryocytic DAMI cell line was transiently tranfected with GFP fused to SDF-1α (SDF-1α-eGFP, for details see supplemental Materials and methods). The cells were stained with fluorescence tagged anti-GFP mAb (green) or anti-vWF mAb (red), respectively, and analyzed by confocal laser scanning microscopy. The overlay (left) shows that SDF-1α-eGFP co-localizes with the α-granular protein vWF. DAMI cells transfected with eGFP (not fused to the C-terminus of SDF-1α, mock-transfected) served as controls (right). Bars, 10 µm.
