(A) Knockdown of UTX in a female mouse embryonic fibroblast cell line that carries the X-linked UTX gene but not the Y-linked UTY gene. Cells were infected with pSUPERretro-based retroviral siRNA construct targeting mouse UTX sequence AGTTAGCAGTGGAACGTTA. After selection with 2 mg/ml puromycin, single colonies were picked and expanded. The expression of UTX was analyzed by Taqman quantitative reverse-transcriptase PCR and normalized with GAPDH level.
