Time-lapse video microscopy recordings of HeLa H2B-GFP cells (Kanda, T., K.F. Sullivan, and G.M. Wahl. 1998. Curr. Biol. 8:377-385) treated with 100 nM Hesperadin. The upper panel shows H2B-GFP, the lower panel H2B-GFP in blue, Nomarski in yellow. The first part shows a cell undergoing mitosis in the presence of Hesperadin. Chromosome condensation appears normal, but chromosomes fail to align to a metaphase plate before the onset of anaphase. Chromosomes fail to segregate in anaphase. Cytokinesis is attempted but not completed. Chromosome movement during anaphase can be better observed in the cell shown in the second part for which the GFP signal was recorded in four Z sections (spaced by 5 µm) that were overlaid to produce the image shown in the upper part.
