Figure S2.Overview of the distribution of Pmel17 and TRP1 in MNT-1 cells. Ultrathin cryosec-tions of MNT-1 cells were double immunogold labeled with HMB50 (PAG 10) and TA99 (PAG 15). (A) Pmel17 is primarily enriched in the lumen of stage II melanosomes (II) where it associates with the internal striations. Pmel17 is also detected in the Golgi apparatus (GA) and electron-lu-cent compartments present near the Golgi apparatus (stars). TRP1 is present in the limiting mem-brane of later stage melanosomes, the Golgi apparatus and in small vesicles near the Golgi appara-tus or melanosomes. (B) Numerous stage IV melanosomes in the cell periphery are intensely labeled with the anti-TRP1 antibody. Bar, 100 nm.
