(A). Total cell lysates from WT (lane 1) or Rsk2 KO (lane 2) mouse spleen T cells were western blotted with antibodies to Rsk1 (clone C21), Rsk2 (clone E1), Rsk3 (clone C20), or Erk1/2 (clone K23). An antibody to actin (clone I19) was used to show equal loading of the proteins. The asterisk indicates cross-reaction of Rsk3 antibody with Rsk2, since it was absent in the lysates from Rsk2 KO spleen T cells. (B) Total cell lysates from 293T+ cells transfected with pRV (lane 1), pRV Rsk1 (lane 2), pRV Rsk2 (lane 3), or pRV Rsk3 (lane 4) were western blotted with antibodies to Rsk1 (C21), Rsk2 (E1), or Rsk3 (C20). An antibody to actin (I19) was used to show equal loading of the protein. The cross reactivity of Rsk3 antibody (C20) with Rsk1 proteins is shown as an asterisk and with Rsk2 proteins as a double asterisk.
