Retinoic acid inhibits Th17 polarization and enhances FoxP3 expression through a Stat-3/Stat-5 independent signaling pathway Blood Elias et al. 111: 1013 Supplemental materials for: Elias et alFiles in this Data Supplement: - Figure S1. ATRA enhances FoxP3 and inhibits IL-17 expression independent of IL-2 (JPG, 92.4 KB) -
Peripheral CD4 cells from Il2−/−RAG2−/−TcR-transgenic mice were polyclonally stimulated for 3 days in media alone or with the addition of exogenous TGF -1, TGF -1 and IL-2 or Th17 conditions (TGF -1, IL-6, anti IFN- and anti IL-4). All stimulations were performed in the presence (lower panels) or absence (upper panels) of 1µM ATRA. IL-17 and FoxP3 expression were measured on fixed cells by intracellular staining.
