Fig. 8. ATL analogs induce vasodilation in isolated rat aorta: relaxation relative to Iloprost. Rats were killed with pentobarbital overdoses. The aorta was isolated and a preload of 300-400 mg was given. The vessels were precontracted with U46619 (25 ng/ml). Relaxation was induced with addition of Iloprost, 5(R/S)-methyl-LXB4, 15-epi-16-phenoxy-LXA4, ATLa1, or ATLa2 to a final concentration of 1 mM. Aorta smooth muscle relaxation was measured with a force transducer, and the data were digitized and stored on a personal computer. 5(R/S)-methyl-LXB4, 15-epi-16-phenoxy-LXA4, ATLa1, and ATLa2 caused 42.8%, 37.0%, 38.1%, and 40.0% relaxation of smooth muscle, respectively. Values represent mean ± SEM (n = 4-6).
