(top) Thymocyte suspensions were stained with anti-CD4 plus anti-CD8 antibody to identify the major thymocyte subsets in individual CSN5/JAB1del/del mice. Shown are representative plots from mice displaying a mild (>30% DP/SP cells; left), moderate (>10 and <30% DP/SP cells; middle), or severe (<10% DP/SP cells; right) phenotype. Percentage values for the various subsets are shown in the respective quadrants. (bottom) Thymic cellularity, as estimated by calculation of the number of thymocytes from individual thymi classified as mild, moderate, or severe after electronic gating of the various subsets. Numbers are the mean ± the SD of values obtained from at least 5 mice per phenotype at 5–6 wk of age. Phenotypes classified as “mild” showed nonetheless a relevant alteration in thymic development compared with WT littermates (Fig. 1).
