B16 melanoma cell before and after treatment with 30 µM blebbistatin. Video corresponds to Fig. 9 D. Cell was transfected with mCherry-actin and GFP-fascin. Note the generation of arc-shaped bundles from microspikes before blebbistatin treatment. Arcs dissolve in blebbistatin but fascin-rich bundles still translocate into lamella. To avoid photoactivation of blebbistatin, fascin images were recorded only as single frames during the video (at 0:00, 12:42, 27:31, and 53:06 min). Time-lapse images were acquired every 15 s and the total duration of the video was 53 min. The display rate is 6 frames/s. Bar, 10 µm.
