Figure S3. 53PB1 chromatin recruitment occurs in PR-Set7–depleted cells. (A) Immunofluorescence of γ-H2A.X and 53BP1 in U2OS cells 3 d after shLuc or shPR7 expression. The nuclear localization of 53BP1 is only observed in shPR7 U2OS cells displaying γ-H2A.X signal, suggesting that 53BP1 is normally recruited to chromatin in response to spontaneous damage in PR-Set7–depleted cells. (B) Percentage of shLuc and shPR7 U2OS cells displaying 53BP1 nuclear localization before or after 10 Gy of ionizing radiation (IR) 2.5 d after shRNA expression. Note the significant increase in the percentage of 53BP1-positive shPR7 U2OS cells after ionizing radiation (compare bar 2 with bar 4), suggesting that 53BP1 is still recruited to chromatin after induced DNA damage in PR-Set7–depleted U2OS cells.
