Figure S3. Differential regulation of IL-12 and IL-23 production in CD1c⁺DCs and monocyte-enriched PBMCs stimulated with H37Rv and LPS. Monocyte-enriched PBMCs (moPBMC) and CD1c⁺ DCs (left) or IFN-γ–primed CD1c⁺ DCs (right) were labeled with [³⁵S]methionine and stimulated with heat-killed H37Rv or LPS, alone or associated with R848. After 18 h, supernatants were immunoprecipitated with anti–IL-12 p35, anti–IL-23 p19, or anti–IL-12/23 p40 mAbs and resolved in nonreducing SDS-PAGE.
