Figure S5. Truncated T-synthase protein in LSC cells is not secreted into the media. LSC and LSB cells stably expressing C-terminal HPC4-tagged T-synthase were treated with 10 µM MG-132 or 0.5% DMSO as a control for 12–14 h. T-synthase from of media (∼15 ml) was absorbed on 200 µl HPC4-Ultralink and the activity was measured as nmol/h-ml beads (A; error bars represent ±1 SD from the mean), and the bound protein was eluted using 5 mM EDTA and analyzed by Western blot (B).
