Dynamic behavior of CD9 in the context of the tetraspanin-enriched compartment. CD9 was labeled with Atto647N-conjugated Fab fragments (red; recorded at 100 ms/frame), and ensemble labeling was performed with the Cy3B-conjugated SYB-1 antibody (green) in PC3 cells. Two examples representative of the CD9 behavior as compared with ensemble labeling are sequentially shown. The first example corresponds to the original data shown in Fig. 2 D. Trapping inside the tetraspanin-enriched area lasted for 60 frames (6 s). The first vidoe is replayed at 100 ms, and the second vidoe is accelerated 2.5×. The pixel size corresponds to 160 nm. White lines correspond to the trajectories of CD9 single molecules tracked in real time.
