Figure S1. GM6001 partially restored spreading of aged human platelets on fibrinogen. (A) Representative flow cytometry dot plots of washed platelets and in vitro–injured platelets in the absence or presence of GM6001. (B) Washed human platelets derived from a healthy volunteer were incubated at 37°C for 24 h. Platelets pretreated with 1% DMSO or 100 µM GM6001 were plated on fibrinogen-coated coverslips in the presence of 1 U/ml thrombin. Representative fluorescence images of platelet spreading are shown. All samples were stained with Alexa 488 phalloidin to mark F-actin (green) and with anti-αIIb antibody followed by Alexa 567 (red). Bar, 10 µm. (C) Spreading was observed using a confocal laser microscope, and surface area was estimated (100 platelets from three independent experiments). Note that GM6001 partially restored the spreading ability of aged human platelets on fibrinogen.
