(D) XRyk MO specifically inhibited the translation of its cognate mRNA. The 5′ untranslated region (500 pg) and ORF (500 pg) XRyk-myc mRNA were injected with XRyk MO (10 ng) into the animal regions of embryos at the four-cell stage. Animal cap explants isolated at early gastrula stages were then subjected to immunoblot analysis with anti-myc antibodies. Actin served as a specificity control. (E and F) Quantitative assays of rescue in Keller sandwich assays (E) and whole embryos (F) were each performed more than three times. n, total number of embryos and explants.
