Figure S1. Immunization and isolation of T naive, T_CM, and T_EM cells. (a and b) C57BL/6 mice were immunized with KLH in CFA, and 4 wk later CD4⁺ T cells were enriched from spleen and lymph nodes and sorted according to the expression of CD44 (a), and CD62L and CD127 (b). Staining before and after sorting is shown. T naive cells were CD44^low, CD62L⁺, CD127⁺; T_CM cells were CD44^high, CD62L⁺, CD127⁺; and T_EM cells were CD44^high, CD62L⁻, CD127⁺. Results are representative of several experiments. Purity was always >98%. Background staining with isotype-matched control antibodies on gated CD4⁺ T cells is shown (a, right). Percentages are shown.
