- Supplemental Figure 1 - Analysis by qRT-PCR confirms T3 regulation obserbed by RT-PCR for TRα (A) and TRβ (B). The RNA samples used in Fig. 2 were analyzed by qRT-PCR as in Fig. 1. Note that the magnitude of induction for TRβ was much bigger than TRα in all organs.
- Supplemental Figure 2 - ChIP assays show that anti-TR (new PB) antibody immunoprecipitated chromatin-bound Xenopus tropicalis TRα (A) and TRβ (B) similarly as anti-TR (PB) antibody (Fig. 4C and D). The experiments were done as in Fig. 4C and D except the use of anti-TR (new PB) antibody. Note that the enhanced signal of the promoter region but not the ampicillin resistance gene in the presence of TR/RXR with our without T3 is similar to that observed with the anti-TR (PB) antibody, suggesting both antibodies recognized TRα and TRβ similarly in the ChIP assay. It is unclear why the background was much higher with anti-TR (new PB) antibody compared to anti-TR (PB) antibody (Fig.4).
