Whole cell lysates from GM6914 (FA-A) fibroblasts expressing Flag-tagged FANCA (lanes 25) or vector control (lane 1) treated with 1mM MMC for 18 hours (lanes 4 and 5) and/or 1mM okadaic acid for 30 minutes (lanes 3 and 5) were immunoprecipitated using anti-FLAG antibody, run on SDS-PAGE and silver stained. The band from lane 5 was excised and analyzed by mass spectrometry. IP: immunoprecipitation.

GM6914 (FA-A) fibroblasts expressing vector only, Flag-tagged wild type FANCA, or FANCA S1449A mutant were treated or not with 1mM MMC for 18 hours. Chromatin extracts were prepared and run on SDS-PAGE and progressively immunoblotted with phosphospecific antibody and anti-FANCA antibody. Both wild type and mutant FANCA localized to chromatin.
