- Data Supplement - Figure S1. Disorazole C1 blocks BrdU uptake in A549 cells. Cells were treated with the 50% growth inhibitory concentrations of discodermolide (DISCO; 25 nM) or vinblastine (VBL; 2 nM), 0.25 2 nM disorazole C1 (C1), 50 nM doxorubicin (DOX), 10 nM O1OA or DMSO vehicle for 24 h and 7 days followed by BrdU uptake, fixing and incubation with fluorescent anti-BrdU antibody (green) and Hoechst 33342 nuclear stain (blue).
- Data Supplement - Figure S2. Disorazole treatment of A549 cells did not produce direct DNA damage. A549 cells were treated with DMSO vehicle (A), disorazole C1, 2 nM (B) or 50 nM (C), doxorubicin, 50 nM (D) or 1 μM (E), vinblastine, 2 nM (F) or 50 nM (G), or UV radiation (60 J/m2) (H). After 4-24 h cells were harvested and the expression of Ser139 phosphorylation on H2AXγ (phospho-H2AXγ) was determined by western blotting with an antibody from Cell Signaling (Danver, MA). GAPDH was used a protein loading control.
