A similar transplant experiment as shown in Fig. 1a was performed except for the fact that a higher dose of normal competitor BMNCs from C57BL/6J (1 × 108, CD45.2+) was co-transplanted with 1 × 107 BMNCs from B6.SJL (CD45.1+) into lethally-irradiated recipients (A). At day 14 after transplantation, BMNCs from leukemic hosts or control were analyzed for the expression of CD45.1, CD150, CD48, c-Kit, Sca-1, and lineage markers with flow cytometry. Representative flow profiles and the average frequency of CD45.1+CD150+CD48−LKS cells in leukemic or control hosts are shown (B). Each value is mean ± s.d.. ** P

BMNCs were stained with lineage antibodies and Annexin-V. Lin− cells were gated by means of a stringent parameter. The representative plot is shown above. The average frequency of the (Annexin V+PI−) fraction in CD45.1+Lin− cells from leukemic or control groups is shown (2 experiments, 5 mice/group in each experiment, P=0.48).
