Fig. S1. CMF70 knockdown does not cause gross disruption of the axoneme. (A,B) The CMF70 knockdown cell line was grown in the absence (A) or presence (B) of tetracycline to induce knockdown. Flagellum skeletons were prepared and subjected to transmission electron microscopy as described in Materials and Methods. Scale bar is 50 nm.
Movies 1-4. CMF70 is required for normal motility and functions as a DRC subunit.
Movie 1: Real-time video of control cells
Movie 2: Real-time video of CMF70 single knockdown. RNAi knockdown of CMF70 causes an erratic and less vigorous flagellar beat, in contrast to the rapid and three-dimensional flagellar beat exhibited by control cells in movie 1.
Movie 3: Real-time video of PF16 single knockdown shows severe flagellar beat defects.
Movie 4: Real-time video of PF16/CMF70 double knockdown. Flagellar paralysis of the PF16 single knockdown (Movie 3) is suppressed, as the flagellum exhibits a more sustained, though not wild type beat.
