Effect of latrunculin A on actin cables in wild-type cells and stable actin mutants. ACT1 and act1-159 cells were grown to mid-log phase and incubated with various concentrations of Lat-A dissolved in DMSO for 5 min. Cells were fixed and stained for F-actin using Alexa-594 phalloidin. The amounts of actin cables were assessed by fluorescence intensity of Alexa-phalloidin bound to F-actin in mother cells that contained actin cables but no actin patches. n > 80 for each Lat-A concentration tested. The curve (Inset) indicates that act1-159 cells treated with 15 µM Lat-A contain the same amount of cable fluorescence as wild-type cells. Error bars represent SEM.
