The kaiA promoter is not specifically essential for generating basic circadian rhythms. kaiA was removed from the wild-type strain by replacement with a kanamycin resistance gene. Then, Ptrc::kaiA was introduced back into the kaiA^- strain (kaiA^-+Ptrc::kaiA). Bioluminescence profiles of the PkaiBC::luxAB reporters in these clones were assayed and displayed as described in Fig. 1.
