Influence of Fos-Jun orientation preference on the synergy between Fos-Jun and NFAT1. (A) Quantitation of the relative efficiencies of transcription activation of the promoters shown in Fig. 4B. The efficiency of transcription activation at each promoter was calculated relative to the unsupplemented reaction, and the ratio between the efficiencies of transcription activation of the two templates indicated below the graph is shown (average of at least 3 independent experiments). A ratio above 1 indicates preferential activation of the promoter linked to the 390 bp transcription unit whereas a ratio below 1 indicates preferential activation of the promoter linked to the 242 bp transcription unit. (B) Absence of preferential transcription of the two templates in the absence of NFAT1. The templates shown in part C were transcribed in the presence of the proteins indicated above the lanes. A long exposure of the transcripts is shown to verify equivalent transcription of both templates in the absence of NFAT1.
