Template dependency of inducible RNA-dependent RNA polymerase (RdRP) activity. Soluble protein extracts were prepared from salicylic acid (SA)-treated tobacco plants and passed through a DEAE column as described (1). The RdRP peak fractions were identified through assays of RdRP activity. The identified peak fractions were pooled and assayed for template dependency of the RdRP activity in absence of added RNA (lane 1) or presence of total RNA isolated from healthy untreated tobacco leaves (lane 2), SA-treated tobacco leaves (lane 3), tobacco mosaic virus (TMV)-infected tobacco leaves (lane 4), or RNA isolated from purified TMV (lane 5).Reference:
