Cell confinement controls centrosome positioning and lumen initiation during epithelial morphogenesis

Video 2.
Actin localization in an epithelial cyst developing in low spreading (high confinement) conditions. MDCK cells stably expressing Life-actin-GFP (gray) were seeded on 700-µm2 collagen I–coated micropatterns to grow cysts and analyzed by time-lapse epifluorescence microscopy with a microscope (AF6000 LX; 40× dry objective). Frames were taken every 10 min for 48 h. After 48 h, DNA was stained with cell-permeable Hoechst at the end of the video recording. Time is given in hours and minutes. Bar, 10 µm.
