The N terminus of SKAP55 enables T cell adhesion to TCR and integrin ligands via distinct mechanisms

JSKAP.SY cells exhibit defective SLP microcluster dynamics and impaired contact stability. J14.SY stably deficient in SKAP55 (JSKAP.SY) but expressing SLP-76.YFP (green) were transfected with control mRFP1 (red) and imaged on glass coverslips coated with 10 µg/ml OKT3. Frames were acquired every 2 s for 5 min by spinning disc confocal microscopy (CSU-10 spinning-disc confocal [Yokogawa], Axiovert 200M stand [Carl Zeiss], 40× Plan-Neofluar oil-immersion lens [Carl Zeiss], and an ICCD camera [Stanford Photonics]).
