Point mutation of the ADAP-binding SKAP55 SH3 domain (W333R) impairs SLP microcluster dynamics and contact stability downstream of TCR ligation. J14.SY expressing SLP-76.YFP (green) transiently transfected with SKAP55.W333R.mRFP1 (red) were imaged on 10 µg/ml OKT3-coated glass coverslips. Frames were acquired every 4 s for 5 min by spinning disc confocal microscopy (CSU-10 spinning-disc confocal [Yokogawa], Axiovert 200M stand [Carl Zeiss], 40× Plan-Neofluar oil-immersion lens [Carl Zeiss], and a CCD camera [Hamamatsu]).
