- Light response of C. elegans expressing WT bovine opsin in neurons. (.wmv, 13.3 MB) - WT opsin worms were incubated with 9–cis–retinal in the dark overnight and transferred to a NGM plate lacking food. Worm motion was recorded under dim light. A flash of blue light (488±20 nm, 1 s, 1000 lux) was applied at the 10 s time point (indicated by the black dot in the top left corner). Paralysis of the worms upon illumination was retained until the end of the movie.
- Light response of C. elegans expressing P23H bovine opsin in neurons. (.wmv, 19.4 MB) - P23H opsin worms were incubated with 9–cis–retinal in the dark overnight and transferred to a NGM plate lacking food. Worm motion was recorded as described in the caption to Supplementary movie 1. P23H opsin worms moved vigorously for 10 s, and this movement was arrested immediately after illumination. In contrast to the WT worms (Supplementary movie 1), complete paralysis was short-lived, as demonstrated by head wiggling and occasional body movement observed as early as 10 s after light exposure.
