Ena/VASP regulates mDia2-initiated filopodial length, dynamics, and function

Supplemental Materials

This article contains the following supporting material:

  • Supplemental Materials
  • Movie01 - Supplementary Movie 1. Movie corresponds to Figure 4A (upper panel). The filopodium that is magnified in the upper panel in Figure 4A is indicated by an arrow. GFP-VASPlabeled filopodia extend persistently from the lamellipodium leading edge of a protruding MVD7 fibroblast. Filopodia never bend or kink.
  • Movie02 - Supplementary Movie 2. Movie corresponds to Figure 4A (middle panel). The filopodium that is magnified in the middle panel in Figure 4A is indicated by an arrow. GFP-mDia2M/A-containing filopodia are characterized by irregular dynamics and do not exhibit directionality: individual filopodia move in and out of focus, and flip backwards on the cell body.
  • Movie03 - Supplementary Movie 3. Movie corresponds to Figure 4A (bottom panels). The filopodium that is magnified in the bottom panels in Figure 4A is indicated by an arrow. Filopodium of MVD7 cell containing GFP-VASP and mCherry-mDia2M/A extends progressively. Notably, GFP-VASP localizes distally to mCherry-mDia2M/A at the tip; mCherry-mDia2M/A is additionally localized along the filopodium shaft. Filopodia that display exclusive mCherry-mDia2M/A labeling exhibit irregular protrusion, kinking, bending and collapsing.
  • Movie04 - Supplementary Movie 4. Movie corresponds to Figure 4E, upper panel. mCherry- VASP-labeled filopodia in MVD7 cells that co-express GFP-zyxin consistently induce formation of nascent focal contacts.
  • Movie05 - Supplementary Movie 5. Movie corresponds to Figure 4E, bottom panel. Filopodia in MVD7 cell co-expressing mCherry-mDia2M/A and GFP-zyxin rarely generate focal contacts.
  • Movie06 - Supplementary Movie 6. Movie corresponds to Suppl. Figure S5. MVD7 cells expressing GFP-VASP form persistently protruding filopodia. GFP-VASP-positive focal contacts in the advancing lamellipodium are formed immediately posterior to these filopodia.
  • Movie07 - Supplementary Movie 7. Movie corresponds to Figure 6B (lower panels). mCherry-EB1 comets (green) continuously protrude into filopodia labeled by GFP-mDia2M/A (red) in MVD7 cells that co-express both proteins. Some EB1 comets even reach the tip complex.
  • Movie08 - Supplementary Movie 8. Movie corresponds to Figure 6B (upper panels). mCherry-EB1 comets (green) move towards the cell periphery in MVD7 cells co-expressing GFP-VASP (red), but do not target VASP-labeled filopodia.
