Complete canthi removal reveals that forces from the amnioserosa alone are sufficient to drive dorsal closure in Drosophila

Supplemental Materials

This article contains the following supporting material:

  • Supplemental Materials
  • Movie04 - Movie 4. Time-lapse confocal micrograph movie of the native embryo in Figure 2B. (ubiquitously expressing DE-cadherin-GFP). The duration of closure captured spans 182 minutes and the time interval between each frame is 2 minutes.
  • Movie03 - Movie 3. Time-lapse confocal micrograph movie of a native embryo in Figure 1B. (ubiquitously expressing GFP-moe-ABD). The duration of closure captured spans 180 minutes and the time interval between each frame is 3 minutes.
  • Movie08 - Movie 8. The corresponding movie of Figure 9B displaying all viewpoints.
  • Movie07 - Movie 7. The corresponding movie of Figure 9A displaying all viewpoints.
  • Movie01 - Movie 1. Time-lapse confocal micrograph movie of the embryo in Figure 1A. (early in dorsal closure ubiquitously expressing GFP-moe-ABD with both canthi removed). The duration of closure captured spans ∼160 minutes and the time interval between each frame is 3 minutes. During laser incision, the interval between each frame is 0.5 sec.
  • Movie05 - Movie 5. Time-lapse confocal micrograph movie of an embryo ubiquitously expressing DE-cadherin-GFP with the anterior canthus removed. The duration of closure captured spans 140 minutes, and the time interval between each frame was originally captured at 10 seconds, but 40 seconds in this movie.
  • Movie06 - Movie 6. Time-lapse confocal micrograph movie of the same embryo as Movie 5. The first 30 minutes after anterior canthus removal was segmented and tracked automatically with our custom software. The time interval between each frame is 10 seconds. Cells with blue traces and numbering are successfully segmented and tracked. Note that the program experienced more difficulty segmenting the smaller cells near the leading edges. A segmentation of this interval (30 min) was used for all Fourier analysis described in detail in the Results section. Any cells with incorrect segmentations upon visual inspection were discarded from any analysis.
  • Movie02 - Movie 2. Time-lapse confocal micrograph movie of the embryo in Figure 2A. (late in dorsal closure ubiquitously expressing DE-cadherin-GFP). The duration of closure captured spans 148 minutes and the time interval between each frame is 2 minutes.
