Supplemental Figure 1.pdf -
Distribution of condensins I and II relative to cyclins and lamin B during the cell cycle. HeLa cells were fixed with the post-extraction protocol, and labeled with anti-cyclin A and anti-hCAP-G/condensin I (A) or with anti-cyclin A and anti-hCAP-H2/condensin II (B). Alternatively, the cells were labeled with anti-cyclin B1 and anti-hCAP-H2 (C) or anti-lamin B and anti-hCAP-H2 (D). Chromosomal DNA was stained with DAPI. The bottom panels show merged images of labeling with anti-G (red; A), anti-H2 (red; B, C and D), anti-cyclin A (green; A and B), anti-cyclin B1 (green: C), or anti-lamin B (green: D). Bars, 10 μm.
Supplemental Figure 2.pdf -
Distribution of Aurora B in condensin-depleted chromosomes. Metaphase chromosome spreads were prepared from control cells or cells depleted of hCAP-E (E-dep), hCAP-G (G-dep) or hCAP-G2 (G2-dep). The chromosomes were stained with DAPI and labeled with anti-Aurora B. Merged images are shown in the bottom. Bar, 2 μm.
