• Supplemental Figure 1.pdf - Cep3p distribution reflects kinetics of CBF3 assembly. Cells containing the GAL1 promoter upstream of 3XHA-CTF13 were cultured in galactose (A), were cultured for five hours in dextrose (B) and then transferred to galactose containing media for 15 (C) and 45 (D) minutes. Cells were fixed and stained with antibodies to Cep3p (red), tubulin (green) and chromosomes stained with DAPI (blue). Following deconvolution, projections of 7-12 0.2μm Z-sections were created. Arrows in panels A and D indicate Cep3p juxtaposed to spindle pole and overlapping with chromosomes. Arrowheads in panels B and C indicate Cep3p populations distinct from spindle poles and non-overlapping with chromosomes. Scale bar is 5 μm.
