Movie 1. The experimental condition included the following. As sample, a mutant a 3b 3g subcomplex (a -C193S, b -His-10 at N terminus, g -S107C/I210C) from a thermophilic Bacillus PS3, was immobilized on a Ni-NTA-coated glass slip and attached to the duplex of a magnetic bead to the g -subunit. The solution used was 200 nM ATP in 50 mM Mops-KOH (pH 7.0), 50 mM KCl, 2 mM MgCl2, and 10% BSA. The time table used was 0-16 s, free active rotation; 26-50 s, ADP inhibition at 9 o’clock (defined as 0° hereafter), 26-29 stall at 0°31-34 stall at +10°, 36-36 stall at + 20°, 41-44 stall at +30°, 47-50 stall at + 40°, activation; and 50 s, active rotation.
