Fig. 7. Fluorescent-labeled confocal image of FITC-labeled oligonucleotide and DAPI-stained nuclei. This representative photomicrograph depicts the cellular uptake of FITC-labeled oligonucleotide. Twenty micrograms of FITC-labeled random oligonucleotide was injected intrathecally by means of lumbar puncture in three rats. The rats were killed at 4, 8, and 12 h after injection. Thirty-micrometer sections were colabeled with the specific nucleus marker DAPI, which is known to form fluorescent complexes with natural double-stranded DNA. The optimal cellular uptake of the FITC-labeled oligonucleotide (full arrow) was observed 8 h after injection in the proximity of nuclei (asterisk).
