Intraclonal correlation between TCR ligand potency and off-rates. T cell clones were stained with the following doses of multimers: clone LAU 156/34, 18 mg/ml A2/tyrosinase_370D multimer and 0.9 mg/ml A2/tyrosinase_370N multimer; clone LAU 132/1D5/1, 0.2 mg/ml A2/tyrosinase_370D multimer and 5.5 mg/ml A2/tyrosinase_370N multimer; and clone LAU 132/1G4/1, 0.3 mg/ml A2/tyrosinase_370D multimer and 2 m g/ml A2/tyrosinase_370N multimer. TCR-multimer dissociation was performed as in Fig. 4.
