(A, B) C57BL/6, BALB/c and C57BL/6 Ifnar1-/- BMDMs were stimulated with B. pseudomallei or LPS for the indicated times. (C) C57BL/6 and C57BL/6 Ifnar1-/- BMDMs were stimulated with LPS for 1 h and treated with Actinomycin D (ActD). (A-C) Il10 mRNA was harvested and quantified by qRT-PCR, normalised to Hprt1 mRNA levels. (D) C57BL/6 and BALB/c BMDMs were treated with 2 or 20 ng/ml IFN-β for 2 h prior to stimulation with B. pseudomallei or LPS for 24 h. IL-10 production was quantified by ELISA.
