Reduction of eNOS activity in transfected EA.hy926 cells. (A) The cellular activities of eNOS in cultured human endothelial EA.hy926 cells transfected with either control-dsiRNA or GLA-dsiRNA were measured based on the biochemical conversion of [3H]L-citrulline from [3H]L-arginine in intact cells. The experiments were performed at day 3 following single GLA-dsiRNA transfection (ST) and day 6 following double GLA-dsiRNA transfection (DT), respectively. (B) Comparison of eNOS activity in GBA-dsiRNA and GAPDH-dsiRNA transfected EA.hy926 cells to control-dsiRNA transfected cells. The experiments were carried at day 6 following double transfections (DT). The enzyme activity was normalized to protein concentration and expressed as pmol/min per mg protein (n = 4).
