Syncytin-1 knockdown-induced BeWo cell apoptosis. (A) Representative results of TUNEL assay (40X). BeWo cells transfected with control (Panel 1) or syncytin-1 specific siRNA (panel 2) were examined with TUNEL for apoptosis at 48 hours post-transfection. Nuclei were counter stained with hematoxylin. Brown color staining in the nuclei indicated cell apoptosis (marked by arrows). (B) Apoptotic and total cells were counted in 10 randomly selected microscopic areas and the percentage of apoptotic cells was calculated for the control (Ctl siRNA) and syncytin-1 knockdown groups (SsiRNA1). A significant increase in apoptotic cells was observed in the syncytin-1 knockdown group compared to the control group (**, P < 0.01). Note the condensed and disintegrated nuclei in some cells following sincytin-1 knockdown.
