CD11c+ DCs were enriched from the spleens of wild-type and CARD9-deficient mice. A and C) 1×105 DC were cultured with 1×105 SM1 T cells in the presence of flagellin (10ng/ml), peptide (6uM) or medium alone for 16h. T cell activation was analyzed by measuring CD69 expression on gated SM1 T cells (CD4+CD90.1+). Upper and lower panel shows the activation of SM1 T cells in the presence of wild-type or CARD9-deficient DCs and C) shows the percentage +/−SEM of CD69+ SM1 T cells cultured with wild-type (open) or CARD9 deficient (filled) DCs. Histograms (B) and bar plots (D) show surface expression of CD25 on gated SM1 T cells cultured with wild-type CD11c+ DCs and the bottom panel represent the activation of T cells with CARD9-deficient DCs. Each histogram is a representative of two individual experiments done in triplicate. Error bar represents mean +/− SEM. P<0.05 (**) showed data are statistically significant.
