(A) Human normal chondrocytes (n=3) were cultured with DMEM in the presence of 10 ng/ml IL-1 or 200 ng/ml TNFα for 0–48 h and LRP1 proteins in the cell lysate and the medium were analysed as in Figure 1A. (B and C) Quantification of LRP1 protein in the cell lysate (B) and sLRP1 released into the medium (C). (D) Effect of metalloproteinase inhibitor CT1746 (20 μM), serine protease inhibitor AEBSF (50 μM) or cysteine protease inhibitor E-64 (10 μM) on cytokine-induced LRP1 shedding.
