Identification of the binding partner by protein thermal shift. Differential scanning fluorimetry assay showing CD154 (CD40L) and not CD40 as the binding partner of our compounds. The presence of 11 (DRI-C2105045, 25 μM; top row) and DRI-C21045 (10 μM; bottom row) caused clear shifts in the melting temperature of the protein for CD154 (right figures; purple vs. blue line), but not for CD40 (smaller insets).
