A working model for the role of SIRT6 in the regulation of YAP/TAZ and hepatic fibrogenesis. When SIRT6 is sufficiently active, YAP and TAZ are suppressed in HSCs. This promotes a quiescent state of HSCs. Under pathogenic conditions, SIRT6 activity is markedly reduced, and this leads to highly acetylated YAP and TAZ that promote HSC activation and fibrogenesis.
