In the article “Membrane-Associated α-Tubulin Is Less Acetylated in Postmortem Prefrontal Cortex from Depressed Subjects Relative to Controls: Cytoskeletal Dynamics, HDAC6, and Depression,” by Harinder Singh, Justyna Chmura, Runa Bhaumik, Ghanshyam N. Pandey, and Mark M. Rasenick, which appeared on pages 4033–4041 of the May 13, 2020 issue, similarities were noticed between Figure 3A and C. The originally published version of this article contained an error in Figure 3C (TX114: Samples Normal 9–12 and Sample Suicide: 24–27) which was a duplicate of Figure 3A (TX114: Samples Normal 1–4 and Sample Suicide: 16–19). The error did not affect numeric results, as the quantitation shown in the paper was carried out with the correct blots, including the one shown below.
Figure 3.
Acetylated tubulin in lipid rafts prepared from postmortem PFC is decreased in depressed subjects relative to control. Plasma membrane was purified, and lipid rafts were prepared by TX-100-resistant (lipid rafts) and Triton X-114-resistant (nonrafts) microdomain isolation. Samples were analyzed as in Figures 1 and 2. A–D, Data for samples isolated from normal controls and depressed suicides. E, Samples isolated from depressed nonsuicides. F, Tubulin and acetylated tubulin were quantified as in Figures 1 and 2. Scatterplots were used to show the extent of tubulin modification. ***p < 0.0001. *The sample lanes in acetylated tubulin and total tubulin are not aligned because the exact same sample was run on two different gels. The stripping of acetylated tubulin blot to reprobe for total tubulin antibody always results in remnants and misleading signal from acetylated tubulin probed blot. Therefore, to ensure that total tubulin is not an artifact match of acetylated tubulin, the same samples were run on different gels and probed with two different antibodies.
The error occurred when a set of JPEG format images derived from original raw blots were transferred to PowerPoint for preparation of the figures. In this process, the blot for Figure 3A, Normal 1–4 and Suicide 16–19 lanes, was mistakenly reused for creating Figure 3C, Normal 9–10 and Suicide 24–27. This error went unnoticed throughout the review process.
In the updated figure, the sample lanes in acetylated tubulin and total tubulin are not aligned because the samples were run on two different gels. Stripping of acetylated tubulin blots to reprobe for total tubulin results in residual signal from acetylated tubulin anti-IgG. Therefore, to ensure that the total tubulin measurement does not contain artifacts due to residual acetylated tubulin, the same samples were run on different gels and probed with two different antibodies.
The corrected Figure 3 and its legend appear below. This error does not affect the conclusions of the paper.
