Most antibiotics are ineffective against Gram‐negative bacteria due to the presence of the outer membrane (OM). In article number 2411923, Jianzhong Shen, Kui Zhu, and co‐workers screened natural xanthone skeletons with the potential to penetrate the OM after amine modification. These optimized compounds cross the OM via a self‐promoted pathway, facilitating the discovery of novel antibacterial agents.
