Revised Highlights.

DNA damage promotes the release of dsDNA from nuclear, mitochondrial, and micronuclear sources, activating the cGAS-STING pathway.

sensing, potentially requiring PTMs and nuclear export.

Intracellular trafficking may control cGAS-STING signaling to balance immune activation and tolerance.

STING agonists are proposed to enhance tumor immunity, but challenges include delivery, toxicity, and context-dependent immune suppression.
