ABSTRACT
The HLA‐A*24:677 allele differs from HLA‐A*24:02:01:01 by a single non‐synonymous nucleotide change in exon 2.

The HLA genes are the most polymorphic group known in the human genome. In this report, we describe a novel HLA‐A allele, designated HLA‐A*24:677, which was identified by Sanger dideoxy nucleotide sequencing of a sample from a Chinese haematopoietic stem cell donor.
Using the DNA Extraction Reagent Kit (Qiagen, Hilden, Germany), genomic DNA was extracted from whole blood. After extraction, the DNA was subjected to HLA genotyping for HLA‐A, ‐B, ‐C, ‐DRB1, ‐DQB1 and ‐DPB1 loci with the specific sequencing primers (CATB, HLA‐SBT Kits, Shanghai, China). Informed consent was obtained from the subject involved.
The HLA‐A*24:677 allele had one nucleotide change when compared to the HLA‐A*24:02:01:01 allele in exon 2 at position 223 where T → G (TGG → GGG) in codon 51, resulting in an amino acid change from Tryptophan to Glycine (Figure 1).
FIGURE 1.

The exon 2 nucleotide sequence of the HLA‐A*24:677 allele compared with the closely related sequence of HLA‐A*24:02:01:01. Dashes (‐) indicate identity with the HLA‐A*24:02:01:01 allele. The HLA‐A*24:677 allele had one nucleotide change when compared to the closely related HLA‐A*24:02:01:01 allele in exon 2 at position 223 where T → G (TGG → GGG) in codon 51, resulting in an amino acid change from Tryptophan to Glycine.
The name HLA‐A*24:677 has been officially assigned by the WHO Nomenclature Committee for Factors of the HLA System in December 2025 (submission number HWS10102016) [1]. This follows the agreed policy that subjects to conditions stated in the most recent Nomenclature Report, names will be assigned to new sequences as they are identified. Lists of such names will be published in the following WHO Nomenclature Report [2].
